Universal viral transport medium
Virus preservation solution is a protective liquid medium in a virus sampling tube. In China, we usually call it Viral transport medium (VTM or UTM) .Generally, PCR detection cannot be directly carried out at the sampling place during nucleic acid detection. We need to collect the swab, and then transfer the sample for inspection. So we need to add VTM kits to collect, transport and store samples.
Virus detection is different from routine biochemical detection. A virus itself is a kind of microorganism with a simple structure, which must parasitize in living cells. After sampling, the virus leaves the host cell, and its protein shell and nucleic acid will degrade quickly in the sampling tube. In this way, it is impossible to judge whether the initially collected sample contains virus during nucleic acid detection, which is easy to cause false negative.
We can solve the above problems by adding a virus preservation solution to the sampling tube:
Universal viral transport medium non inactivated and inactivated.
1. viral transport medium inactivation type. This mixed solution medium contains high-efficiency virus lysate, which can quickly inactivate the virus after sampling, effectively prevent the risk of secondary infection. And release virus nucleic acid for easy detection.
2. The VTM solution contains nuclease RNase inhibitor, which can prevent the nuclease in the cytoplasm from degrading the viral nucleic acid after the degradation of broken cells in the sample, protect the stability of viral nucleic acid to a greater extent, and greatly improve the nucleic acid extraction efficiency.
3. It has good tightness, which is convenient for storage and transportation and saves transportation cost.
Different from the above-inactivated viruses, the non inactivated virus preservation solution does not contain lysate.
It is a variety of solution components conducive to the culture of virus-host cells, such as Hanks solution base, gentamicin, fungal antibiotics, bovine serum albumin (V), cryoprotectant, biological buffer and amino acids.
This VTM can ensure the integrity of the virus and help to increase the survival time and infection stability of the virus.
Therefore, it has higher requirements for the protection of virus detection and sample storage temperature after sampling.
Non inactivated virus preservation solution can be used for the collection and transportation of many specimens, including a variety of viruses and mycoplasma, Ureaplasma, chlamydia and so on.
Whether inactivated or non inactivated virus preservation solution, its basic function is to protect the integrity of virus or virus nucleic acid and ensure the accuracy of virus nucleic acid detection. The two VTMS have their own advantages.
